ABSTRACT

Marine fungi are important members of the marine microbiome, which have been paid growing attention by scientists in recent years. The secondary metabolites of marine fungi have been reported to contain rich and diverse compounds with novel structures (Chen et al., 2019). Aspergillus terreus, the higher level marine fungus of the Aspergillus genus (family of Trichocomaceae, order of Eurotiales, class of Eurotiomycetes, phylum of Ascomycota), is widely distributed in both sea and land. In our previous study, the coral-derived A. terreus strain C23-3 exhibited potential in producing other biologically active (with antioxidant, acetylcholinesterase inhibition, and anti-inflammatory activity) compounds like arylbutyrolactones, territrems, and isoflavones, and high sensitivity to the chemical regulation of secondary metabolism (Yang et al., 2019, 2020; Nie et al., 2020; Ma et al., 2021). Moreover, we have isolated two different benzaldehydes, including a benzaldehyde with a novel structure, from A. terreus C23-3 which was derived from Pectinia paeonia of Xuwen, Zhanjiang City, Guangdong Province, China.

ABSTRACT

Objective Investigate the clinical features,diagnosis and treatments of the scimitar syndrome,and different forms of treatment to alleviate pulmonary hypertension.Methods A retrospective analysis of clinical data of 14 children with scimitar syndrome from 2013 to 2017,including clinical symptoms and signs,chest X ray,echocardiography,chest CT and cardiac catheterization,treatment outcome and follow-up.Assess embolization of systemic pulmonary collateral and pulmonary venous drainage correction surgery,which is better for lowering pulmonary blood flow.Results 14 patients with scimitar syndrome were diagnosed from 2013 to 2017.There were 5 boys and 9 girls;3 cases <7 kg in weight.Scimitar syndrome was suspected because of extroversion,and diagnosed by color Doppler echocardiography and 13 of them confirmed by cadiac CT scan when ascimitar vein was detected entering the inferior vena cava.11 patiens had right lung dysplasia and 4 had horseshoe lung.Three patients had severe pulmonary arterial hypertension,3 had moderate to severe pulmonary arterial hypertension,and 2 had moderate pulmonary arterial hypertension,the left had slight pulmonary arterial hypertension.4 patients had pulmonary venous drainage correction surgery,after that 2 of them had systemic pulmonary collateral embolism.6 patients systemic pulmonary collateral embolism first,then 4 of them had surgical repair,1 case of 13 years old asymptomatic child without surgery.1 patient with heart failure,severe pulmonary hypertension,pulmonary infection,died before surgery,while another died after surgical repair.At last 1 patient was lost for follow-up visits.Systemic pulmonary collateral embolism and pulmonary venous drainage correction surgery could all reduce blood flow of pulmonary.After systemic pulmonary collateral embolism,patients had slight pulmonary arterial hypertension just need follow-up visits.Conclusion Clinically,found children with heart of dextrocardia position,growth retardation,recurrent lung infections,unexplained right heart failure,pulmonary hypertension,should consider the possibility scimitar syndrome.Whether pulmonary vascular embolization or surgical repair,can significantly reduce pulmonary artery's blood flow and alleviate pulmonary hypertension to protect pulmonary,even reduced the incidence of pneumonia and mortality.So we suppose ealy pulmonary hypertension in scimitar syndrome patients maybe dynamic pulmonary hypertension.

ABSTRACT

Objective To explore the accuracy and practicability of the three-dimensional ultrasound in real-time intellectual positioning navigation.Methods The phantom was applied in the real-time intellectual positioning guided biopsy of three dimensional ultrasound,free-hand ultrasound-guided biopsy and bracket-guided biopsy with different depths(5 cm,10 cm) and different sizes of the targets (6 mm and 15 mm).The time spent,the length of the sample and the success rate of biopsy were compared among three groups,and statistical analysis was done.Results ①The different sizes of targets with different depths were shown in three kinds of biopsy that the real-time intellectual positioning guided biopsy of three-dimensional ultrasound spent more time than bracket-guided biopsy,less than free hand ultrasound-guided biopsy.The difference was significant (P <0.05).The aquired red-dyed sample was longer in the real-time intellectual positioning guided biopsy of three-dimensional ultrasound compared to free-hand ultrasound-guide biopsy and bracket-guided biopsy,the difference was significant (P <0.001).②For real-time IP-guided biopsy of three-dimensional ultrasound with same depth,larger size of targets(15 mm) spent less time than smaller saize of targets (6 mm),the difference was significant (P < 0.05).However,compared to smaller saize of targets(6 mm),success rate of biopsy in larger size of targets(15 mm) had no significant difference (P =0.238 at depth of 5 cm,P =0.479 at depth of 10 cm).③For real-time intellectual positioning guided biopsy of three-dimensional ultrasound with same size,time spent was less in the targets at the depth of 5 cm than in the targets at the depth of 10 cm,the difference was significant (P<0.05).The aquired red-dyed sample was longer in the targets at the depth of 5 cm than in the targets at the depth of 10 cm,and success rate of biopsy in the targets at the depth of 5 cm was higher too (P <0.05).Conclusions The real-time intellectual positioning guided biopsy of three dimensional ultrasound spend less time with high accuracy and success rate of biopsy,especially for smaller and deeper targets,can be clinically applied with accuracy and practicability.

ABSTRACT

Objective To screen the genes most relevant to lymph node metastasis of cervical cancer and identify the genes at the key knots of the regulatory network to provide the potential targets for cervical cancer intervention.Methods The transcriptional profiling database of TCGA was used, and random forests algorithm was adopted to rank the genes related to lymph node metastasis extracted from GeneCards database.STRING and Cytospace tolls were used to build the interactive regulatory network and identify the most weighted genes localized in the central of the network.DAVID platform was used to perform a functional annotation for the whole geneset.Results We ranked 2784 genes in respect to their potential contributions to lymph node metastasis of cervical cancer and identified the genes at the key knob.The genes related to cancer metastasis were enriched to cytokines pathway, MAPK pathway, wnt pathway, intercellular interaction, adhesive conjunction, cellular skeleton regulation, etc.Some of the identified key genes, like EGFR, NOTCH1, RHOA, etc. have been verified to be closely related cervical cancer metastasis in the basic and clinical research. Conclusion Random forests algorithm is useful, taking advantages of TCGA database, in enriching the genes playing significant role in cervical cancer metastasis.A majority of the genes in the analyzed geneset were indicated to be significantly correlated with lymph node metastasis.

ABSTRACT

Advanced and recurrent cervical cancer has a poor prognosis and few effective therapeutic option.With the development of molecular biology,the targeted drugs for patients with locally advanced cervical cancer continuously appear,such as inhibiting human papillomavirus,targeting angiogenesis,epidermal growth factor receptor,mammalian target of rapamycin,cyclooxygenase-2,sarcoma gene,proteasome inhibitor,DNA methylation,etc.With the development of new molecular targeted therapy,more effective or even treatment measures of cervical cancer might be found,which can provid a new approach for the individualized treatment of cervical cancer.

ABSTRACT

Objective:To investigate the efficacy and safety of metformin in the treatment of elderly patients with diabetes. Meth-ods:Totally 72 cases of elderly patients with diabetes in our hospital from January 2012 to December 2012 were randomly divided into the observation group and the control group. The observation group was received metformin, and the control group was given repaglin-ide. The efficacy and safety of the two groups were compared. Results:After the treatment, blood sugar of fasting, 2h after meals and 10:00 p. m. in the observation group and the control group was significantly decreased (P0. 05). The HbA1c levels before and after the treatment in the observation group was with statistically significant differ-ence (P0. 05). The incidence of adverse reactions in the observation group was lower than that in the control group(P<0. 05). Conclusion:Effect of metformin in the treatment of elderly patients with diabetes is remarkable and safe, which is worthy of further promotion and application.

ABSTRACT

Objective:To discuss the effect of amlodipine combined with atorvastatin in the treatment of elderly patients with acute cerebral infarction hypertension. Methods:Totally 86 cases with acute cerebral infarction hypertension were randomly divided into the treatment group (43 cases) and the control group (43 cases). The control group received amlodipine 5mg, po, qd, and the treatment group was given atorvastatin 20mg additionally, po, qd. After the one-month treatment, the efficacy of the two groups was studied and compared, and the changes in neural function defect scale, blood pressure, blood lipids and CRP were also investigated. Results:Af-ter the treatment, the total effective rate and the total effective rate of antihypertensive effect in the treatment group were both signifi-cantly higher than those in the control group (P0. 05), and those in the treatment group was significant lower than those in the control group (P<0. 05). Conclu-sion:Amlodipine combined atorvastatin in the treatment of elderly patients with acute cerebral infarction hypertension shows significant effect with low incidence of adverse drug reaction, which is worthy of wider application.

ABSTRACT

Objective This study aimed to study the effect of intra-brachial nitroglycerin administration via the angiography catheter on radial artery spasm in transradial percutaneous coronary intervention. Methods From patients undergoing transradial percutaneous coronary intervention (TRI) and succeeding in coronary angiography (CAG), 810 patients who needed percutaneous coronary intervention (PCI) (including Percutaneous Transluminal Coronary Angioplasty and/or stent implantation) were erolled, including 413 patients in the study group and 397 patients in the control group. When coronary angiography completed, positioned the angiography catheter over brachial artery and patients in the group received an administration of 400μg nitroglycerin via the angiography catheter. The control group received an administration of 400μg nitroglycerin via the sheath. The incidence of RAS were recored and compared. Resultes Baseline characteristics were similar between two groups. In the study group, the incidence of RAS was signiifcatively lower (18 vs. 33, P=0.021). Conclusions Intra-brachial nitroglycerin administration via the angiography catheter can prevent RAS during the insertion of guiding catheter in TRI.

ABSTRACT

Objective: Construction mouse GM-CSF gene effective eukatyotic expressive vector, selection high GM-CSF expressing mouse leukemia cell line RMA after transfected with the constructed vector, and study the method of treatment leukemia with tumor cells transfected with GM-CSF gene.Methods:770 bp of GM-CSF 3' end cDNA was amplified by PCR and inserted into pcDNAS vector.The constructed vector was transfected into RMA cells by electroporation. After screening by G418 and cloning by limiting dilution, a relative high GM-CSF expressing cell clone was selected by RT-PCR, hematopoietic progenitor cell proliferative assay and hematopoietic progenitor cell colony formation assay. The cells of this clone were inactivated by mitomycin-C and vaccinated mice to induce antitumor immune reaction. Results: The orientation and sequence of the insert was found to be correct, and a GM-CSF high expressing cell line RMA-GM was selected , which can induce mice obtain anti-tumor protective immune ability after inactivated by mitomycin-C.Conclusion:Tumor cells transfected with GM-CSF gene may be used an effective anti-T lymphycoma tumor vaccine.

ABSTRACT

Objective: To clone human T cell receptor (TCR) ? chain gene and express its encoding protein by baculoviral expression system in insect cells. Methods: TCR ? chain cDNA was cloned from normal human peripheral blood mononuclear cells (PBMC) by RT-PCR and inserted into baculoviral transfer vector. This vector was co-transfected with baculovirus into insect sf 9 cells. The recombinant protein expressed was identified by SDS-PAGE and flow cytometry with mouse anti-human ? chain monoclonal antibody. Results: Human TCR ? chain cDNA cloned and inserted into baculoviral vector specifically expressed protein in the insect sf 9 cells, accounting for about 11% of the total protein yield in the supernatant of cell lysate. The molecular weight of the recombinant protein determined by SDS-PAGE was identical to what we anticipated. The insect cells transfected with recombinant baculovirus were demonstrated by intracellular labeling flow cytometry to express ? chain protein. Conclusion: Human T cell receptor ? chain gene was successfully cloned from human PBMC, and its encoding protein was highly expressed in insect cells. The TCR ? chain protein obtained by bioengineering technique is useful for in depth biological function study.
